Water-for-Injection loop extended to the second filling line

The site's Water-for-Injection loop has been extended to serve the second aseptic filling line, with the whole distribution network revalidated rather than only the new branch.

Extending a water loop changes the hydraulics of the entire system: flow velocity, temperature profile and sanitisation coverage all shift. Treating the extension as a self-contained addition would have left the original branches unproven under the new conditions, so requalification covered the complete network.

You cannot extend a water loop and requalify only the new pipe. The hydraulics do not respect the boundary.

WFI is generated by distillation and held in hot circulation, monitored for conductivity, total organic carbon, bioburden and endotoxin, with every user point sampled over a defined cycle. Endotoxin control is the parameter that matters most for a parenteral product, and it is trended rather than merely tested.

The loop's performance forms part of the annual review for every sterile product it touches.
